Fenbendazole, a benzimidazole anthelmintic agent, exerts its therapeutic effects primarily by selectively binding to β-tubulin in parasitic cells, disrupting microtubule polymerization and leading to impaired glucose uptake, glycogen depletion, and subsequent breakdown of cellular integrity and function, resulting in anthelmintic activity without significant impact on mammalian hosts due to lower binding affinity in human tubulin.
This mechanism also extends to potential anticancer applications by destabilizing microtubules in tumor cells, activating the p53 tumor suppressor pathway, inhibiting GLUT1 glucose transporters and hexokinase activity to starve cancer metabolism, and modulating multiple cellular pathways for apoptosis induction, showing preclinical promise in reducing tumor growth and metastasis in models of colorectal, lung, and lymphoma cancers while its broad antiparasitic spectrum covers roundworms, hookworms, whipworms, Taenia species, and Giardia for comprehensive management of gastrointestinal and systemic infections.
